Using polymerase chain reaction (PCR), a specific segment of DRB gene was amplified using crude extracts of DNA from frozen blood; DR specificities were types with nonradioactively-labeled oligonucleotide probes.
Abstract
The sample consisted of heparinized blood from 8 families with 58 members. The samples were washed with saline twice after removal of plasma and frozen for eight years; they had been typed for HLA antigens previously. The amplified products clearly detected the DRB gene segment. The results agreed with the serological typing and clearly distinguished each specificity. This study demonstrates that DNA analysis can be performed with frozen blood samples; it should be possible to type HLA class II antigens in field population studies. 11 references.
