This study explored the effectiveness of Whatman FTA cards at extracting and storing DNA from the Calliphoridae (Diptera).
Abstract
Recently forensic entomology has turned to the use of insect DNA to provide identification of insects for time of death estimations. The use of insect DNA requires that the DNA be extracted efficiently and intact for use in molecular procedures and for long-term storage. Whatman FTA cards have been utilized for DNA collection and processing; the current study evaluated the ability of these cards to produce high quality DNA samples suitable for use with polymerase chain reaction (PCR). The criteria under examination were the overall ability to be amplified, the potential for use with a variety of insect life stages, and the ability to amplify products of considerable size to ensure samples are not degraded during the extraction process. Results showed DNA samples that were able to be amplified from a variety of life stages. Whatman FTA cards thus produced DNA samples of adequate quality and quantity for use in PCR procedures.
